Will this kit allow for "stacking" of single-DIN units, i.e., a single-DIN stereo and a DIN-sized CB radio?

Maybe .. it really depends on the units you plan on installing. Remember, must single-DIN units mostly worry about the face size, not the body size.
With all the cutting needed to get it in, it might be rough.

After installation, will the A/C vents still be functional, and does it come with the vent directional controls installed, or do you have to transfer the old ones to this unit?

Yes, the vents will still work after the swap as before assuming you transfer them correctly. They have snaps on both sides. Firm pressureon the edges and they will pop out or in.
You also have to move the metal retention snaps from the old to the new trim bezzle. Those are harder as they press fit. Take your time and gently use players that fit. Per-share for a good snap fit as needed.
To install a double din radio, which is the goal of this, you also have to cut the vent ducting inside the dash opening in two places to clearance the radio. This is the slow and tedious part. Measure well. It's better to take too little than too much. Once clearanced, you cover the openings in the vent ducts with the provided aluminium ducting tape. Instructions are provided. Be sure to clean all the trimming particles out well first.
Once it's all in it looks factory. One note though. If you are 6ft tall or over like I am you can no longer see the symbols at the top of the environmental controls without bending to the right a bit due to the new bezzle contours. You likely have them memorized but I pass it in anyway. I have found it a small trade off.

Not sure of specific display size, but any double-Din unit should fit, you will just need to trim back more of the inner dash and ductwork. I used it for an alpine ilx-w650 and it worked great!

It's a double dinn which is a set standard for the size of the head unit itself, whatever size display the manufacturers can fit Within those limits is up to them.

The instructions are clear. All that has to be done it removing the old vents and retaining clips from the bezel and install them on the new one. And to use the driver-side bracket you need to cut some of the plastic behind the dash. Overall very little work for a big improvement.
